Determination of chlorophyll and mangiferin content in mango leaves by using UV-VIS spectrum.

Abstract : Objective: UV-VIS Spectrum was used to quantitatively analyze the content of mangiferin and chlorophyll in mango leaves in order to provide references for comprehensive exploitation and utilization of Guangxi mango leaves resources in future. Method: Light absorption values of mangiferin and chlorophyll in mango leaves were measured using UV-VIS Spectrum, and then the contents of mangiferin and chlorophyll were conversed based on linear regression equation and Arnon formula. Meanwhile, the contents of mangiferin and chlorophyll in 11 kinds of mango leaves of Baise in Guangxi were studied. Result: The best wavelength for measuring mangiferin standard substance and the content of mangiferin in extract of mango leaves was 258.5 nm. When adopting UV-VIS Spectrum to determinate mangiferin content, the linear relationship (R2=0.9998) of the regression equation (y=0.0697x-0.0009) was good within the scope of 4.12-41.20 µg/mL. The method was accurate with good repeatability and the test results were stable. The contents of mangiferin and chlorophyll in 11 kinds of mango leaves of Baise in Guangxi ranged from 11.493 to 16.675 µg/mL. The content of Red ivory was low, while that of Taiya and Nongyuan No. 5 was higher. The total chlorophyll content ranged from 4.83 to 8.77 mg/mL. The contents of Guifeihong and Chiinhuang were comparatively high, while those of the Sijimi and Nongyuan No.5 were low. Conclusion: UV-VIS Spectrum could be used to evaluate the medicinal material quality of mango leaves.
