Abstract : Anthocyanin synthesis is a main metabolic approach for red color development in mango fruit. Phenylalanine ammonialyase (PAL) is the last enzyme in the anthocyanin synthesis. PAL catalyzes the instability proanthocyanidin to anthocyanin. Degenerate primers were designed according to the reported PAL gene sequences, and the full-length cDNA sequence of PAL gene of mango fruit was obtained with 3? RACE and 5? RACE. The open reading frame of the gene was 2160 bp in length, encoding 719 amino acids with the molecular weight of 79.18 kD. A 2200 bp fragment was amplified from its genome, and it did not contain introns. Phylogenetic analysis found that the gene encoded protein had a close relationship with that from Horsetail, Fir, Ginkgo, et al. The expression of the PAL gene in different mango varieties showed that PAL was highly expressed in 'Guifei' varieties, while lowly expressed in green Guiqi varieties.
