Short-term conservation of mango seed.

Abstract : Seeds of mango (Mangifera indica) cultivars Alphonso and Dashehari were soaked in 1% 8-hydroxyquinoline sulfate for 10 min then packed immediately into polythene bags and stored at 5°, 15° or room temperature (25-35°C). In a second experiment, stones and kernels were stored separately at 5°C; germination was blocked in Alphonso kernels by application of ABA for 30 min. Seeds were treated with a cryoprotectant (10% glycerol and 10% DMSO) for 30 min and stored under similar conditions to seeds in the first experiment. Samples were removed periodically and germinated at 30°C, and data on seedling vigour, length and dry weight were recorded in 21-day-old seedlings. Seed viability of both cultivars was significantly affected by storage time and temperature with the best results obtained at 15°C (50% viability at 12 months). None of the seeds treated with either ABA or cryoprotectants survived after 4 weeks of storage. However, longevity of seeds of both cultivars was extended from 2 weeks to 12 months by pretreatment with 8-hydroxyquinoline sulfate at 15°C.
