Abstract : A 2-step protocol was developed for in vitro rooting of microshoots excised from plantlets obtained from nucellar somatic embryos of mangoes cv. Amrapali. Role of auxins, their concentrations, treatment periods and culture illumination conditions for conferring competence for rooting were ascertained. IBA was the most potent among the 3 auxins (IBA, IAA and NAA) tested for promoting efficient rooting and root growth. The best rooting (89.71%) was observed on auxin-free agar-gelled rooting medium after a 24 h pulse treatment with 5 mg IBA/litre in liquid root induction medium in dark. Dark conditions favoured root induction and root growth; incubation in the light (16 h photoperiod) was inhibitory.
