Callus induction and root initiation from explants of zygotic embryos in mango hybrid.

Abstract : Zygotic hybrid mango embryos resulting from a cross between cv. Amrapali and Sensation were excised from young fruitlets (60-day-old) and germinated aseptically. Explants (epicotyl segment, leaf petiole and shoot tip) from the germinated embryos (120 days) were excised and cultured to note their in vitro response. The maximum callus formation (90.3%) was recorded with epicotyl segments cultured on half strength Murashige and Skoog's medium containing 500 mg/l casein hydrolysate+200 mg/l glutamine+60 g/l sucrose+200 mg/l PVP and supplemented with a growth regulator combination of 2,4-D (2.0 mg/l)+BAP (0.5 mg/l). Direct root organogenesis was also noted in epicotyl and shoot tip cultures incubated in dark in the medium containing low level of 2,4-D alone.
