Abstract : A simplified procedure involving extraction of homogenized tissue in 1.0 M sodium acetate buffer followed by 48 h dialysis against distilled water has been standardized for the assay of polygalacturonase and cellulase activities of mango (cv. Totapuri), tomato (cv. Arka Vikas) and pawpaw (cv. Surya). Satisfactory enzyme activities could be obtained by using a one-step extraction procedure without adjusting pH and overnight stirring. This procedure could, therefore, be used effectively to process a large number of samples.
