Cloning and sequence analysis of RGAs genes encoding serine/threonine protein kinase-like (STK) proteins from mango (Mangifera indica).

Abstract : A polymerase chain reaction was performed with genomic DNA extracted from peak-green leaves of mango cv. Zihuamang, using the degenerated primers designed according to the conserved amino acid sequences of catalytic domains I and IX of STK proteins. Six 500bp DNA fragments were amplified and identified as resistant gene analogues (RGAs) by sequence alignment in the Genbank. The molecular phylogenic tree analysis of the 6 fragments and 2 RGA genes (Xa21 from rice and Pto from tomato) also showed they were RGAs. The 6 fragments were named as PK1-PK6 genes, respectively. The proteins encoding by PK3, PK4 and PK6 genes had all the catalytic domains of STK proteins. In addition, the proteins encoding by PK1-PK6 genes showed high analogies with receptor-like protein kinase.
