Genetic similarity analysis among Mangifera indica cultivars/lines based on DNA markers amplified by RAPD and specific NBS primer.

Abstract : The objective of this study was to analyze the nucleotide binding leucine-rich repeat (NBS-LRR) in analog resistance gene family in Mangifera indica using randomly amplified polymorphic DNA (RAPD) markers. The combinations of 15 RAPD primers and 1 NBS specific primer were used to amplify NBS sequences complementary to the DNA sequences from 32 cultivars/lines. Eighty-three polymorphic bands of the NBS-LRR resistance gene analogs were obtained. The data were then analyzed by unweighted pair-group method using arithmetic averages (UPGMA) and neighbor joining (NJ) programs. Five groups were clustered ranging from 0.64 to 0.84 of similarity coefficient, indicating that NBS might be used for phylogeny of the analog resistant gene in mango. Some specific polymorphic bands obtained in this study could be used for mango cultivar identifications.
